Epithalon (also spelled Epitalon) is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) developed by Professor Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology. It is a Khavinson bioregulator, a class of short peptides that regulate gene expression in specific tissues at very low concentrations. Epithalon targets the pineal gland and activates telomerase, the enzyme responsible for maintaining telomere length on chromosomal ends. Telomere shortening is one of the primary hallmarks of cellular aging. Epithalon is the synthetic analog of Epithalamin, a peptide extract originally derived from bovine pineal gland. It is administered in short, intensive courses (10-20 days) rather than continuously, consistent with Khavinson's bioregulatory dosing philosophy.

Often run 1-2 times per year. Total cycle dose is 100mg. (Alternative protocol: 5mg daily for 20 days). Human use has been published, but almost entirely by a single research group in Russian-language journals. None of it is registered on any trial registry and most reports were unblinded, so this evidence is far weaker than a controlled trial.

How it works

Epithalon activates the catalytic subunit of telomerase (hTERT) in somatic cells, enabling the enzyme to add telomeric repeats (TTAGGG) to chromosomal ends. This counteracts the progressive telomere shortening that occurs with each cell division and is one of the recognized hallmarks of aging. Epithalon also normalizes pineal gland function, restoring melatonin production patterns that decline with age. Melatonin itself has antioxidant and immunomodulatory properties. As a Khavinson bioregulator, Epithalon works through gene expression regulation at very low concentrations, interacting with DNA at the level of histone modification and transcription factor accessibility rather than through traditional receptor binding. This mechanism explains why bioregulators are administered in short intensive courses followed by extended rest periods.

What to expect

Early
Days 1–7

Days 1-10 (during course): Sleep quality improvements may be noticeable (pineal/melatonin normalization). Gene expression changes are initiated but not yet manifested clinically. Course is only 10-20 days.

Mid
Weeks 2–4

Weeks 2-12 (after course): Gene expression changes continue to manifest. Telomerase activation effects persist beyond the dosing period. Sleep improvements may sustain.

Later
Weeks 4–12

Months 3-12: Cumulative effects across multiple courses. Telomere length measurements (if tracked) may show stabilization or modest lengthening over time. Best assessed through longitudinal biomarker tracking.

Epithalon induced telomerase activity and telomere elongation in telomerase-negative human fetal fibroblasts.

Addition of Epithalon peptide in telomerase-negative human fetal fibroblast culture induced expression of the catalytical subunit, enzymatic activity of telomerase, and telomere elongation, which can be due to reactivation of telomerase gene in somatic cells and indicates the possibility of prolonging life span of a cell population and of the whole organism.

Claims we could not support

No abstract we checked supports these for this molecule at this route. That is not the same as saying they are false — it marks where the evidence is missing.

Dose/route: 5 mg SubQ daily for 10-20 days, 2-3x/year. No checked abstract administered this regimen.

Pineal/melatonin and epigenetic mechanisms. The checked abstract addressed telomerase only.

Primary safety and malignancy concern. No human SubQ abstract established tolerability or cancer risk.

04

The honest take

Telomerase activation

Supported

Confirmed in multiple cell culture studies showing hTERT activation and telomere elongation. The mechanism is well-supported. Whether this translates to meaningful anti-aging effects in humans requires long-term data.

Lifespan extension
Supported

Animal studies show approximately 25% lifespan increase in rodents. One of the few compounds with lifespan data. Not confirmed in humans.

Pineal / melatonin restoration

Animal data shows normalization of melatonin production in aged animals. Consistent with Khavinson's bioregulatory theory of organ-specific gene expression restoration.

Interactions & stacking

Two different questions: what is documented about running these together, and what is documented about drawing them into the same syringe. A good pharmacological partner can still have to be pinned separately.

Pin alone. Epithalon must not share a syringe with anything else, whatever the pharmacology says. PIN ALONE. Khavinson bioregulators work through gene expression regulation at extremely low effective concentrations. Mixing with other peptides could interfere with the precise signaling mechanism.

Documented together

SS-31Epithalon targets telomere maintenance (cellular lifespan), SS-31 targets mitochondrial membrane integrity (cellular energy). Two fundamental hallmarks of aging addressed simultaneously. Pin separately.
PinealonBoth target pineal gland through different bioregulatory mechanisms. Can be run in sequence for extended pineal support.

Keep separate

GHK-CuCopper ions from GHK-Cu degrade Epithalon's peptide structure. Never mix in the same syringe. Separate injection sites recommended when dosed close in time.

Check any specific pair in the Stack & Interaction Checker, which answers both questions separately. Absence of a documented conflict is not evidence of safety.

06

Side effects & safety

Commonly reported

  • Very well tolerated
  • Mild injection site irritation
  • No significant side effects reported in published literature

Less common & notes

  • Epithalon is a small tetrapeptide with a clean safety profile in published research.
  • The short course duration (10-20 days) limits cumulative exposure.
  • Theoretical concern about telomerase activation in cancer cells, though animal studies actually showed reduced tumor incidence.
  • Individuals with active malignancy should consult their oncologist before use.

Storage. Store unreconstituted vials frozen (-20C) or refrigerated (2-8C). After reconstitution with BAC water, refrigerate at 2-8C. Stable for 28-30 days. Small tetrapeptide with favorable stability. Short 10-day course means reconstituted vial is typically used within 5-10 days, well within stability window.
