Pomegranate (Punica granatum) is a fruit native to the Middle East and South Asia, used medicinally for over 5,000 years across multiple cultures. Pomegranate contains an exceptionally high concentration of polyphenols, especially punicalagins (the largest molecular weight polyphenols known) and ellagitannins, which are converted by gut bacteria into urolithins - particularly urolithin A. Urolithin A has emerged as one of the most exciting longevity compounds in recent years for its ability to stimulate mitophagy (selective clearance of damaged mitochondria).

How it works
Pomegranate's primary active compounds are punicalagins (large polyphenols) and ellagitannins, which are hydrolyzed in the gut to ellagic acid and then converted by specific gut bacteria (especially Gordonibacter species) to urolithins - particularly urolithin A. Notably, only 30-40% of people have the gut bacteria to efficiently convert ellagitannins to urolithin A; this 'urolithin metabotype' partly explains inter-individual variability in pomegranate response. This led to the development of direct urolithin A supplementation (Mitopure) which bypasses the bacterial conversion step.
Urolithin A's most important mechanism is induction of mitophagy - the cellular process of selectively clearing damaged mitochondria. With age, accumulation of damaged mitochondria contributes to reduced cellular energy efficiency and various age-related dysfunctions. Urolithin A reactivates mitophagy through PINK1/Parkin pathway activation, leading to improved mitochondrial quality and function. Animal studies show urolithin A extends lifespan and improves muscle strength in aged mice.
Beyond urolithin A, pomegranate's whole polyphenol profile provides direct antioxidant effects (especially against oxidized LDL, which drives atherosclerosis), eNOS upregulation (improved endothelial function and erectile function), 5-alpha reductase inhibition (relevant for prostate health), and modulation of inflammatory pathways (NF-kB inhibition). Punicic acid in seed oil has additional effects on lipid metabolism and PPAR-gamma signaling.
