THE POPULAR CLAIM IS NOT WHAT THE STUDY SHOWED. At 10 mg/day for 4 weeks in 18 healthy men, plasma ESTRADIOL rose significantly (51.9 to 73.9 pmol/L, p<0.004) while testosterone showed only a non-significant trend. The 3 mg/day work raised both, but only relative to an artificially boron-DEPLETED diet, which is not a normal baseline.

How it works

Boron's hormonal effects are mediated through multiple pathways. The most striking is reduction in sex hormone binding globulin (SHBG) - the protein that binds testosterone and estradiol in blood, rendering them inactive. By lowering SHBG, boron increases the free fraction of these hormones available for tissue action. The mechanism may involve modulation of hepatic SHBG synthesis or direct effects on hormone-binding protein turnover.

Boron also directly affects steroidogenesis by upregulating expression of testicular enzymes including 17-beta hydroxysteroid dehydrogenase, 17-alpha hydroxylase, and aromatase. The net effect in men is increased free testosterone with smaller relative increases in estradiol. In postmenopausal women, boron raises estradiol levels (potentially supporting bone density) without raising estrone or estriol disproportionately. Boron has also been shown to inhibit phosphodiesterase enzymes, contributing to increased nitric oxide signaling and improved endothelial function.

Beyond hormones, boron is essential for the function of multiple boron-dependent enzymes including some involved in bone metabolism (alkaline phosphatase modulation), joint cartilage health, and antioxidant defense (NAD+ pool support). Boron also has direct anti-inflammatory effects via modulation of interleukin and TNF-alpha pathways, and may inhibit acetylcholinesterase mildly (cognitive support mechanism). The pleiotropic activity makes boron broadly metabolic-supportive rather than narrowly hormonal.

Mechanism/effect boundary: in a small pilot (n=8 healthy men), acute and one-week oral supplementation with 10 mg/day boron increased plasma boron concentration and was associated with changes in steroid hormones (increased free testosterone, decreased estradiol) and reduced inflammatory biomarkers (hsCRP, TNF-alpha); this is a small, non-placebo-controlled preliminary study, not a validated dose-response finding.
