Humanin is a 24-amino acid mitochondrial-derived peptide (MDP) encoded by the 16S rRNA gene of mitochondrial DNA. It was originally discovered in 2001 from surviving neurons in Alzheimer's disease brain tissue, suggesting a role in cellular survival under stress. Humanin is cytoprotective, anti-apoptotic, and neuroprotective, activating the STAT3 signaling pathway through a trimeric receptor complex (CNTFR/WSX-1/gp130). Along with MOTS-c, Humanin represents a class of signaling peptides that communicate mitochondrial health status to the rest of the cell and to distant tissues. Humanin levels decline with age, correlating with increased cellular vulnerability.

How it works

Humanin binds to a trimeric receptor complex (CNTFR/WSX-1/gp130), activating JAK-STAT3 signaling which upregulates anti-apoptotic genes (Bcl-2 family) and suppresses pro-apoptotic signals (Bax, caspases). It also binds intracellularly to IGFBP-3 and Bax, directly blocking apoptotic cascade initiation. Humanin provides cellular stress resistance by maintaining mitochondrial membrane potential, reducing ROS production, and preventing cytochrome c release. As an MDP, it functions as a retrograde signal from mitochondria to nucleus, communicating mitochondrial health status and activating protective gene expression programs.
