How it works

PQQ's primary mechanism is activation of mitochondrial biogenesis through the PGC-1alpha pathway. PQQ stimulates phosphorylation of CREB (cAMP response element-binding protein), which activates PGC-1alpha transcription and protein expression. PGC-1alpha is the master regulator of mitochondrial biogenesis, driving expression of nuclear-encoded mitochondrial proteins, NRF1/NRF2 (nuclear respiratory factors that activate mitochondrial DNA replication and transcription), and downstream mitochondrial biogenesis machinery.

PQQ also functions as a redox cofactor capable of 20,000+ catalytic cycles - this catalytic efficiency makes PQQ a particularly long-lived antioxidant. It scavenges superoxide, hydroxyl radicals, and peroxynitrite, protecting mitochondria from oxidative damage during high-energy-demand states. The combination of biogenesis stimulation AND antioxidant protection is mechanistically unique - PQQ both creates new mitochondria and protects them.

Additional mechanisms include induction of nerve growth factor (NGF) expression (supports neurite outgrowth and neuronal survival), modulation of cellular signaling pathways including AMPK and AKT, anti-inflammatory effects via NF-kB inhibition, and supportive effects on glucose homeostasis. PQQ also crosses the blood-brain barrier and accumulates in mitochondria-rich tissues including brain, heart, skeletal muscle, and liver.
