SAMe (S-Adenosyl-L-Methionine) is a methyl donor cofactor present in every cell, derived from methionine. It is the body's primary methylation agent, donating methyl groups for DNA methylation, neurotransmitter synthesis, phospholipid metabolism, and many other reactions. Unlike most supplements, SAMe is FDA-approved as a prescription drug in many EU countries (for depression and osteoarthritis) but sold as a dietary supplement in the US.

How it works

SAMe is the universal methyl donor in the cell:

Methylation Reactions

SAMe donates methyl groups to DNA, proteins, phospholipids, neurotransmitters, and many small molecules. Converts to SAH (S-adenosylhomocysteine), then back to homocysteine and methionine via methylation cycle.

Neurotransmitter Synthesis

Required for synthesis of catecholamines (dopamine, norepinephrine) and serotonin precursor reactions.

Phospholipid Metabolism

Supports neuronal membrane phospholipid synthesis (phosphatidylcholine via methylation of phosphatidylethanolamine).

Joint Cartilage

Stimulates proteoglycan synthesis in chondrocytes - basis of osteoarthritis indication.

Liver Glutathione

Methionine cycle intersects with cysteine and glutathione synthesis - supports hepatic detoxification.

Bioavailability Critical

SAMe is unstable and rapidly degraded by stomach acid. Enteric coating or butanedisulfonate stabilization is essential for absorption.

Claims we could not support

No abstract we checked supports these for this molecule at this route. That is not the same as saying they are false — it marks where the evidence is missing.

Dose/route: 200-400mg via Oral enteric-coated tablet, 1-3x daily. No checked live PubMed abstract supported this complete molecule/formulation/route/dose/frequency combination.

Primary safety claim: Mild stimulation / anxiety (early dosing). No checked live abstract directly established this as the primary concern for the card's exact formulation and regimen.

Side effects & safety

Commonly reported

  • Mild stimulation / anxiety (early dosing)
  • Insomnia (PM dosing)
  • Mild GI upset

Less common & notes

  • MANIA in bipolar patients - critical contraindication.
  • L-DOPA antagonism in Parkinson's.
  • Allergic reactions rare.
  • Long-term safety good per EU pharmaceutical use.
