UC-II Collagen (Undenatured Type II)

UC-II · Undenatured Type II Collagen

UC-II is a patented form of undenatured type II collagen extracted from chicken sternum cartilage using a specific low-temperature, non-enzymatic process that preserves the native triple-helix structure of collagen molecules. This is fundamentally different from hydrolyzed collagen peptides (which are denatured into amino acid fragments) - UC-II works via a unique 'oral tolerance' immune mechanism rather than providing collagen building blocks.,The mechanism: small daily doses (40mg) of undenatured type II collagen interact with Peyer's patches in the small intestine, training the immune system to recognize joint cartilage type II collagen as 'self' rather than as a target for autoimmune attack. This downregulates the inflammatory response against joint cartilage that drives osteoarthritis and rheumatoid arthritis progression.,UC-II has demonstrated superior efficacy vs glucosamine + chondroitin in head-to-head RCTs for osteoarthritis. Despite the dramatically smaller dose (40mg/day UC-II vs 1,500mg/day glucosamine + 1,200mg chondroitin), UC-II produced greater pain reduction and functional improvement. The 40mg dose is critical - higher doses do NOT provide more benefit (and may even reduce the oral tolerance effect).

Take on empty stomach (morning before food) for optimal Peyer's patch exposure. The triple-helix collagen structure is sensitive to gastric acid, so empty stomach increases delivery to small intestine. Single daily dose - higher doses do NOT help and may reduce effect.

How it works

UC-II works through 'oral tolerance' (also called mucosal tolerance) - a fundamental immune mechanism in which exposure to small amounts of an antigen via the gut trains the systemic immune system not to attack that antigen elsewhere in the body. This is the opposite of vaccination logic.,When undenatured type II collagen is consumed daily in small doses (40mg), it is sampled by M cells overlying Peyer's patches in the small intestine. The collagen is presented to T-regulatory cells (Tregs) in gut-associated lymphoid tissue (GALT). These Tregs then circulate systemically and, when they encounter type II collagen in joint cartilage, they suppress local inflammatory T-cell responses (Th1, Th17) and promote tolerance.,The result is reduced autoimmune attack on joint cartilage, decreased pro-inflammatory cytokine production (TNF-alpha, IL-1, IL-17) at the cartilage interface, and reduced cartilage matrix degradation by MMPs (matrix metalloproteinases).,Critical: this mechanism requires NATIVE (undenatured) collagen with intact triple-helix structure. Hydrolyzed collagen peptides lose the structural epitopes that Tregs recognize, so they cannot trigger oral tolerance. They have different (smaller, building-block) effects.,Critical: dose matters - 40mg/day is the validated therapeutic dose. Higher doses may shift the immune response from tolerance to active recognition (the original mechanism behind some RA studies actually used much higher doses to attempt to provoke immune responses, demonstrating the dose-dependence).

Interactions & stacking

What's safe to combine, and what belongs in a separate pin.

Safe & synergistic

Hydrolyzed Collagen PeptidesDifferent mechanism - peptides provide building blocks, UC-II triggers oral tolerance. Compatible to use both for comprehensive joint approach. Don't expect peptides to replace UC-II's function or vice versa.
Glucosamine/ChondroitinCompatible but UC-II generally outperforms glucosamine/chondroitin in head-to-head trials. Some users layer both.
Hyaluronic Acid (oral)Both joint-targeted via different mechanisms - UC-II for immune tolerance, HA for synovial fluid and joint lubrication. Common joint stack.
BoswelliaAnti-inflammatory complement to UC-II's immune modulation.

Keep separate

Immunosuppressive medications (theoretical)UC-II's mechanism modulates immune function (induction of Tregs). Theoretical interaction with immunosuppressants - discuss with prescriber if on chronic immunosuppression.
Active immunization period (theoretical)Theoretical concern that oral tolerance modulation could affect immune response to vaccines. Likely not clinically meaningful but worth noting.

Pharmacokinetics

Illustrative plasma concentration · 24 h
Half-life
Not applicable in traditional sense - UC-II works locally in gut, not systemically absorbed. Effects are downstream (Treg induction).
Peak · Tmax
Not applicable - oral tolerance effects build over days-weeks, not hours.
Elimination
Not relevant - UC-II is digested in gut. Tolerance effects (Treg-mediated) persist for weeks after dosing stops.
Activity
Tolerance effects build over 60-90 days of daily dosing. Effects persist 4-8+ weeks after stopping (Treg lifespan).

Storage. Store at room temperature (15-25C) in a cool, dry place. Protect from heat and humidity. Capsules shelf-stable 2+ years.

Regulatory status

Classified as a dietary supplement in the US - no prescription required. Widely available. UC-II is a registered trademark of Lonza (formerly InterHealth). No FDA-approved medical indication.
