Snap-8 (Acetyl Octapeptide-3) is a cosmetic peptide developed by Lipotec (now Lubrizol) as a topical anti-wrinkle ingredient. The 8-amino-acid sequence (Ac-Glu-Glu-Met-Gln-Arg-Arg-Lys-Trp) is engineered to inhibit SNARE complex formation in facial muscle cells - mimicking the mechanism by which botulinum toxin (Botox) blocks acetylcholine release at neuromuscular junctions, but at much lower potency and via direct SNARE inhibition rather than enzymatic cleavage.

How it works

Snap-8 inhibits SNARE complex formation, mimicking botulinum toxin's mechanism but via a much weaker pathway:

SNARE Complex Function

Synaptosomal-Associated Protein (SNAP-25) is one of the SNARE proteins required for vesicle fusion at neuromuscular junctions. In facial muscles, SNARE-mediated acetylcholine vesicle fusion drives muscle contraction and wrinkle formation over time.

SNAP-25 Mimicry

Snap-8's 8-amino-acid sequence shares a portion of the SNAP-25 N-terminal sequence. The mimic peptide competes with native SNAP-25 for SNARE complex assembly, partially blocking complete SNARE complex formation.

Reduced Acetylcholine Release

With incomplete SNARE complex formation, acetylcholine vesicle fusion at neuromuscular junctions is reduced - muscle contraction force decreases, repeated contractions create less wrinkle formation.

Topical Skin Penetration

Snap-8 has moderate skin penetration but does not reach deep muscle layers. The cosmetic effect is primarily on superficial expression lines, not deep muscle activity.

Dramatic Potency Difference vs Botox

Botulinum toxin enzymatically cleaves SNAP-25 (irreversible at the receptor), producing dramatic and lasting (3-6 month) muscle paralysis from a single injection. Snap-8 is a competitive mimic (reversible) requiring continuous topical application for modest effect. Magnitudes of effect differ by orders of magnitude.

Methionine-Containing

Sequence contains Met - chemistry conflict with copper-conjugated peptides like GHK-Cu in formulation contexts (less relevant for topical use where formulation chemistry can manage this).
