Kisspeptin is a peptide encoded by the KISS1 gene that acts as the master upstream regulator of the reproductive hormone axis. It stimulates GnRH (gonadotropin-releasing hormone) neurons in the hypothalamus, which in turn triggers LH and FSH release from the pituitary, driving testosterone production and spermatogenesis. Kisspeptin acts upstream of GnRH, making it the most physiologically natural way to stimulate the entire HPG (hypothalamic-pituitary-gonadal) axis.

How it works

Kisspeptin binds to the KISS1R (GPR54) receptor on GnRH neurons in the hypothalamus, triggering GnRH pulsatile release. This cascade stimulates pituitary LH and FSH secretion, which drives testicular testosterone production and spermatogenesis. Unlike HCG (which bypasses the hypothalamus/pituitary by directly stimulating Leydig cells), Kisspeptin preserves the entire HPG feedback loop.

Confirmed. Kisspeptin is the most upstream activator of the reproductive axis, preserving all natural feedback mechanisms.
