Glucosamine is an amino sugar (2-amino-2-deoxy-D-glucose) naturally produced in the body and a precursor for glycoproteins, glycosaminoglycans, and proteoglycans - the structural macromolecules of cartilage and connective tissue. How it works

Glucosamine acts through multiple mechanisms:

Cartilage Substrate

Glucosamine is incorporated into glycosaminoglycans (GAGs) and proteoglycans that form cartilage matrix. Supplementation provides substrate for chondrocyte synthesis.

Anti-Inflammatory

Modulates IL-1β-induced inflammation in chondrocytes; reduces NF-κB activation. Mechanism for symptomatic OA improvement beyond simple substrate effect.

Hyaluronic Acid Synthesis

Supports synovial fluid HA - improves joint lubrication.

Sulfate Form Specificity

Glucosamine sulfate provides bioavailable sulfate for GAG synthesis - explains why sulfate form has stronger clinical evidence than HCl form.
