TWO SAFETY ITEMS. (1) The 1989 eosinophilia-myalgia syndrome outbreak was traced to a manufacturing contaminant rather than tryptophan itself, but it killed and disabled users - product quality matters here more than for most supplements. (2) Serotonin syndrome risk when combined with SSRIs, MAOIs or other serotonergic agents, including Methylene Blue. On food: take with carbohydrate rather than protein - carbohydrate lowers competing large neutral amino acids and improves transport into the brain, while a protein meal does the opposite.

How it works

L-Tryptophan is upstream of 5-HTP in the serotonin pathway:

Tryptophan Hydroxylase

TPH converts L-tryptophan to 5-HTP (rate-limiting step). Then AADC converts 5-HTP to serotonin.

Slower, More Physiologic

TPH-rate-limited conversion produces gentler serotonin elevation than direct 5-HTP supplementation.

Melatonin Pathway

Serotonin → melatonin in pineal gland - sleep support mechanism.

Competing for Brain Entry

L-Tryptophan competes with branched-chain amino acids (BCAAs) for transport across BBB. Carbohydrate-rich snack increases insulin, lowering BCAAs and improving tryptophan brain entry - basis of 'turkey makes you sleepy' folklore.
