Beta-Alanine is a non-proteinogenic amino acid (β-alanine vs the standard α-alanine in proteins) that combines with histidine to form carnosine in muscle tissue. Carnosine is a major intramuscular pH buffer that delays muscle fatigue during high-intensity exercise (60-240 second efforts where lactic acid accumulation is rate-limiting).

How it works

Beta-Alanine works through chronic muscle carnosine elevation:

Carnosine Synthesis

β-Alanine combines with histidine via carnosine synthase to form carnosine. β-Alanine is rate-limiting; histidine is abundant.

pH Buffering

Carnosine is the major intramuscular pH buffer at physiological pH. During high-intensity exercise, lactic acid accumulation lowers muscle pH; carnosine buffers H+ ions.

Time-To-Fatigue

Higher muscle carnosine delays the pH drop that drives fatigue, extending time-to-failure in 60-240 second efforts (most weight room sets, sprint intervals, CrossFit WODs).

Loading Required

A single dose has minimal acute effect. 4-8 weeks of daily 4-6g loading raises muscle carnosine 50-80%; effect persists weeks after discontinuation.

Maintenance Dose

1-2g daily can maintain elevated carnosine after loading phase.

Paresthesia

Bolus β-Alanine causes histamine release producing tingling/itching sensation lasting 30-60 minutes. Harmless but uncomfortable. Split-dosing (800mg-1.6g per dose, multiple times daily) avoids this. Sustained-release formulations also reduce paresthesia.
